I dosed one of my does (not pregnant) with Ivermectin today and in the struggle to do it the lock came off of the tube and she ended up getting 3x more than she needed. Is this a problem?

More than likely she will be fine. Most does are lower than what they should be anyway. As long as she's not pregnant she should be fine. I would just keep an eye on her.

It has a pretty high safety margin. You shouldn't have any issues.

This happened to our neighbour too with a doe when he used his new drench gun for the first time...it didn't cause any problems.
