I've banded with and without using banamine.
I personally always feel better knowing I did the best I could to relieve any discomfort. It does hurt, and will for about 8 hours or so.

Just make sure you give a tetanus shot either before you band or when you do it.
This is crucial, to prevent your animal from contracting tetanus. We didn't one year when we banded calves and lost 2 of them to tetanus. It was a very costly oversight that we will never do again!

I second the idea of making sure you do something to control flies.

Make sure you feel both testicles in the scrotum before you release the band.
